Usually, Yosemite refers to the national park.

Accorditing to the dictionary, the definition is as follows.

So if somebody says'"Let's go to Yosemite." Does "Yosemite" mean "Yosemite Valley" or "Yosemite National Park"?

It will mean both, since the valley is part of the park. Here in California, where I live and where the Park is, we say, "We went to Yosemite for the weekend." We mean the Park, which includes the valley, where the Yosemite River runs through the Park.
Yosemite National Park is named for the river valley, so they are synonyms in common usage.
