Dona is Mrs. Josefina is Josephine.
Actually the word "doña" comes from the Latin word "domina" which means lady.

In Portuguese this title is equivalent to Mrs or Maam and placed before a namne to indicate respect.

In Spanish, the term is "honorific" or a courtesy title or form of address for a woman, placed in front of her first name.

Doña Josefina = the title of respect plus " Josephine." (Note that I did not translate "doña."
