Question

When trying to purify a protein using column chromatography, what happens to the mass of the protein eluted from the column if you accidentally run it dry? Would it be more or less? Thanks.

My best guess is that it would be less. If the column runs dry the protein sticks to the stationary phase, and you have no assurance that (a)the protein will re-enter the mobile phase in its entirety, and (b) that no degradation of the protein will occur.
