Do you think it's possible to use "speak" in the following context: "Speak on the role of the monarchy" or "Speak on the major challenges facing the country at present" (a question at an exam or something like that)?

In oral exams, yes.

In written exams, it'd be better to use the verb "discuss" or "analyze" or "explain," among many others.
