Grade 11 learners want to investigate which enzyme is responsible for chemical digestion of starch into glucose in humans. They are provided with two different enzymes ( carbohydrase and protease),starch powder, warm water at about 37 degrees Celsius and basic apparatus and chemicals to test for glucose .
1.1 select and arrange the method steps in the correct sequence from the list below:
-allow apparatus to stand for an hour
-make two solutions of the same amount of starch, with warm water in test tube 1 and 2
-add 5ml carbohydrase to test tube 1 and 5ml protease to test tube 2
-Place the test tubes in a dark cupboard.
-Add Benedict's solution or Fehling's A +B,in the test tubes to test for glucose.
-Place the test tubes in a warm bath.
-if the contents change from light blue to green /yellow /orange in the test tube, then the enzyme has digested starch to glucose and the hypothesis must be accepted for the chosen enzyme.
-Make two solutions of different amounts of starch ,with warm water in test tube 1 and 2.
-If the contents remain light blue in the test tube ,then the enzyme has not digested starch to glucose and the hypothesis must be rejected for the chosen enzyme.
