Basically, I think it's referring to the choices we make in life. Sometimes we're faced with two distinct directions to choose from, and we can choose only one. But now speculate: What would have happened if you had made a different choice at some point?
Do you think Frost is speculating his choice in the 4th stanza? Does he sigh because he was happy with his decision to take the road less traveled?
That's for each of us to interpret for ourselves. One thing about Frost's work (as with all good poetry) is that each reader's interpretation is quite individual.
