Books' titles need to be in italics or underlined.
I have read the book, <i>The Secret</i>.

1 and 2 mean about the same thing. 3 means the same, but adds emphasis that the speaker knows the content of the book well. It would be better to write #3 like this, though:
I have read the book, <i>The Secret</i>, and I know its content.
I have read the book, <i>The Secret</i>, and I know its content.
