does it make a diffrence what two points you chooose on a stragight line to find the slope of the line

From an algebraic point of view, no. From a practical point of view - if you're trying to measure the height and horizontal distance with a ruler, then yes - because the errors will be greater the closer the points are together. But I think you mean the former situation, not the latter.
